ABSTRACT

Abstract The in vitro sporulation of Didymella bryoniae is of great importance for studies that require pure inoculum and in large quantities. Thus, the objectives of this study were to identify the best condition for D. bryoniae sporulation combining different light spectra (UV-A or UV-B light, white light, and continuous dark), with distinct culture media (PDA, V8, ML, and PDAB) and, to evaluate fungus' survivability stored at -20°C over time. The fungus samples were only able to sporulate when subjected to the UV-B light treatment, regardless of the culture medium. The highest appearance of spores conidium type was observed in the PDAB medium, and the lowest production occurred in the ML medium. Reproductive structures, such as perithecia and pycnidia, were observed in all culture media. However, there was considerable variation in the amount of each structure between the different culture media. The ML and V8 media showed a greater number of perithecia and the PDA and PDAB media presented a greater proportion of pycnidia compared to perithecia. The storage duration at -20°C did not affect mycelial growth or mycelial growth rate. In conclusion, the UV-B light is essential for D. bryoniae in vitro sporulation. Moreover, the culture medium composition influences the type of fungal structure produced, as well as spores' size and quantity. Freezing at -20°C is an efficient technique that can be used to store D. bryoniae for at least five months without loss of viability.


Resumo A esporulação de Didymella bryoniae in vitro é de grande importância para estudos que requerem inóculo puro e em grandes quantidades. Assim, os objetivos deste estudo foram identificar a melhor condição para esporulação de D. bryoniae combinando diferentes espectros de luz (luz UV-A ou UV-B, luz branca e escuro contínuo) com distintos meios de cultura (PDA, V8, ML e PDAB) e, avaliar a sobrevivência do fungo armazenado a -20°C ao longo do tempo. As amostras de fungo só esporularam quando submetidas ao tratamento com luz UV-B, independentemente do meio de cultura. Maior aparecimento de esporos do tipo conídio foi observado no meio PDAB, e a menor produção ocorreu no meio ML. Estruturas reprodutivas, como peritécios e picnídeos, foram observadas em todos os meios de cultura. No entanto, houve uma variação considerável na quantidade de cada estrutura entre os diferentes meios de cultura. Os meios ML e V8 apresentaram maior número de peritécios e os meios PDA e PDAB apresentaram maior proporção de picnídeos em relação aos peritécios. A duração do armazenamento a -20°C não afetou o crescimento micelial ou a taxa de crescimento micelial. Em conclusão, a luz UV-B é essencial para a esporulação de D. bryoniae in vitro. Além disso, a composição do meio de cultura influencia o tipo de estrutura fúngica produzida, bem como o tamanho e a quantidade dos esporos. O congelamento a -20°C é uma técnica eficiente que pode ser usada para armazenar D. bryoniae por pelo menos cinco meses sem perda de viabilidade

ABSTRACT

Abstract Calvatia is a genus of gasteroid fungi, comprising about 47 species worldwide. In this paper we report the second worldwide occurrence of two poorly known species of Calvatia, recorded in the Cerrado biome of Brazil: C. oblongispora and C. nodulata. Detailed morphological descriptions and illustrations, including scanning electron micrographs of hyphae and basidiospores are provided, as well a discussion on their taxonomy and geographic distribution.


Resumo Calvatia é um gênero de fungos gasteroides que compreende cerca de 47 espécies em todo o mundo. Neste artigo relatamos a segunda ocorrência de duas espécies pouco conhecidas de Calvatia, registradas no bioma Cerrado do Brasil: C. oblongispora e C. nodulata. Descrições morfológicas detalhadas e ilustrações são fornecidas, incluindo micrografias eletrônicas de varredura de hifas e basidiósporos, bem como uma discussão sobre sua taxonomia e distribuição geográfica.

ABSTRACT

Abstract Colletotrichum is one of the most economically important fungal genera, which affects a wide range of hosts, specifically tropical and subtropical crops. Thus far, there have been several records of mycovirus infection in Colletotrichum spp., primarily by viruses of the Partitiviridae family. There have also been records of infections by mycoviruses of the Chrysoviridae family. Mycoviruses are (+)ssRNA and dsRNA genome viruses, which may or may not be enveloped. To date, no mycovirus with a DNA genome has been isolated from Colletotrichum spp. Typically, mycoviruses cause latent infections, although hypo- and hypervirulence have also been reported in Colletotrichum spp. In addition to its effects on pathogenic behavior, mycovirus infection can lead to important physiological changes, such as altered morphological characteristics, reduced vegetative growth, and suppressed conidia production. Therefore, research on mycoviruses infecting phytopathogenic fungi can help develop alternative methods to chemical control, which can cause irreversible damage to humans and the environment. From an agricultural perspective, mycoviruses can contribute to sustainable agriculture as biological control agents via changes in fungal physiology, ultimately resulting in the total loss of or reduction in the virulence of these pathogens.


Resumo Colletotrichum é um dos gêneros fúngicos mais importantes economicamente, afetando uma ampla gama de hospedeiros, especialmente em cultivos tropicais e subtropicais. Atualmente já existem diversos registros de infecção por micovírus em Colletotrichum spp., sendo a maioria dos já identificados classificados na família Partitiviridae. Ocorrem registros também de micovírus pertencentes à família Chrysoviridae. Compreendem vírus de genoma de (+)ssRNA e dsRNA que podem ser ou não envelopados. Ainda não foram identificados micovírus com genoma de DNA isolados de Colletotrichum. A infecção por micovírus pode ocorrer de forma latente, mas já foi observado em Colletotrichum spp. o fenômeno de hipo e hipervirulência. Além de influenciar no comportamento patogênico, a infecção pode causar mudanças fisiológicas importantes como alterações das características morfológicas, redução do crescimento vegetativo e redução na produção de conídios. O estudo com micovírus em fungos fitopatogênicos traz uma alternativa ao controle químico que é um método capaz de causar danos irreversíveis ao homem e o meio ambiente. Sob a perspectiva agrícola, os micovírus podem contribuir para agricultura sustentável como agentes de controle biológico. Isso porque obsevam-se mudanças importantes na fisiologia fúngica resultando na perda total ou redução da virulência desses patógenos.

ABSTRACT

In the digital transformation of Chinese pharmaceutical industry, how to efficiently govern and analyze industrial data and excavate the valuable information contained therein to guide the production of drug products has always been a research hotspot and application difficulty. Generally, the Chinese pharmaceutical technique is relatively extensive, and the consistency of drug quality needs to be improved. To address this problem, we proposed an optimization method combining advanced calculation tools(e.g., Bayesian network, convolutional neural network, and Pareto multi-objective optimization algorithm) with lean six sigma tools(e.g., Shewhart control chart and process performance index) to dig deeply into historical industrial data and guide the continuous improvement of pharmaceutical processes. Further, we employed this strategy to optimize the manufacturing process of sporoderm-removal Ganoderma lucidum spore powder. After optimization, we preliminarily obtained the possible interval combination of critical parameters to ensure the P_(pk) values of the critical quality properties including moisture, fineness, crude polysaccharide, and total triterpenes of the sporoderm-removal G. lucidum spore powder to be no less than 1.33. The results indicate that the proposed strategy has an industrial application value.

ABSTRACT

It is unclear how VvLaeA functions in regulating the growth and development of Volvariella volvacea (Bull. ex. Fr.) Sing.. Firstly, bioinformatics analysis of VvLaeA was carried out in this study. Subsequently, the Vvgpd promoter and the open reading frame (ORF) fragment of VvlaeA were amplified and fused by polymerase chain reaction (PCR). The fusion fragment was cloned into the pK2 (bar) plasmid. The recombinant construct pK2(bar)-OEVvlaeA was transfected into Beauveria bassiana by Agrobacterium tumefaciens-mediated transformation. Finally, the growth and development of the transformants were examined. The results showed that VvLaeA shared a low homology with similar proteins in other fungi. Compared with the wild type, the colony diameter of the transformant was significantly increased. However, the pigment deposition, conidial yields and germination rates were significantly decreased. The overexpression strains were more sensitive to stresses than that of the wild type. Further studies showed the conidial cell wall properties of the transformants were altered, and the expressions of genes related to the conidial development were significantly down-regulated. Collectively, VvLaeA increased the growth rate of B. bassiana strains and negatively regulated the pigmentation and conidial development, which shed a light for the functional identification of straw mushroom genes.

ABSTRACT

Coffee is the most appreciated beverage in worldwide; Brazil is the largest producer and exporter of this commodity. Organochlorine endosulfan was banned from the country in 2013 due to its teratogenic agent-related features. Since then, coffee plantations have experienced increased Hypothenemus hampei infestation rates. The aim of the current study is to assess variations in the rates of Coffea arabica fruits brocaded by H. hampei after the application of entomopathogenic fungal species Beauveria bassiana IBCB66. Experiments were carried out with 'Catuaí' and 'Mundo Novo' cultivars between 2018 and 2020, during the borer transit period. Three experiments were carried out based on the application of the aforementioned fungal species on the investigated coffee plant species, both by spraying and sprinkling, at 30-day intervals; 10 fruits were collected per face of each useful plant in each repetition. The experiment has followed a randomized blocks design with five treatments, including the control, and five repetitions, each. Beauveria bassiana Ecobass (IBCB66) wettable powder spray, at the concentration of 2 × 1013·ha­1, was used in experiments I and II. On the other hand, the mix used in experiment III was prepared with blastospores at concentration of 5 × 1012·ha­1 blastospores + 0.1% Silwet. The sprinkling process in all three experiments has used dry aerial conidia at concentration of 2 × 1013·ha­1. Collected data were subjected to analysis of variance (ANOVA), which was followed by Fisher's test at 5% probability level, in the SISVAR software. More than 35,000 fruits were assessed. In addition to variations between experiments, results have evidenced that the rate of brocaded fruits remained high.

ABSTRACT

Brown rot is a common disease in the cultivation and production of Gastrodia elata, but its pathogens have not been fully revealed. In this study, the pathogenic fungi were isolated and purified from tubers of 77 G. elata samples with brown rot. Pathogens were identified by the pathogenicity test and morphological and molecular identification. The pathogenicity of each pathogen and its inhibitory effects on Armillaria gallica were compared. The results showed that 119 strains of fungi were isolated from tubers of G. elata infected with brown rot. Among them, the frequency of separation of Ilyonectria fungi was as high as 42.01%. The pathogenicity test showed that the pathogenicity characteristics of six strains of fungi were consistent with the natural symptoms of brown rot in G. elata. The morphological and molecular identification results showed that the six strains belonged to I. cyclaminicola and I. robusta in the Nectriaceae family of Sordariomycetes class, respectively. Both types of fungi could produce pigments, conidia, and chlamycospore, and the growth rate of I. cyclaminicola was significantly higher than that of I. robusta. The comparison of pathogenicity showed that the spots formed by I. cyclaminicola inoculation were significantly larger than those of I. robusta inoculation, suggesting I. cyclaminicola was superior to I. robusta in pathogenicity. The results of confrontation culture showed that I. cyclaminicola and I. robusta could signi-ficantly inhibit the germination and cordage growth of A. gallica. A. gallica also inhibited the growth of pathogens, and I. cyclaminicola was less inhibited as compared with I. robusta. The results of this study revealed for the first time that I. cyclaminicola and I. robusta were the pathogens responsible for G. elata brown rot.

ABSTRACT

Abstract Microsporidia are obligate intracellular fungi with a remarkable ability to infect a wide range of invertebrate and vertebrate hosts. Namely, Enterocytozoon bieneusi is the most frequently microsporidia reported worldwide, and mainly associated with chronic diarrea and wasting syndrome in AIDS patients. Microscopy and PCR-based detection techniques are effective for diagnosis and identification of species and genotypes; however, these methods should be standardized in each laboratory. In this study, we performed microscopy and nested PCR techniques with PCR product sequencing to detect E. bieneusi in human stool samples. These techniques, if applied together, might prove useful for diagnosis and future epidemiological studies of intestinal microsporidiosis in Argentina.


Resumen Los microsporidios son hongos intracelulares obligados con una notable capacidad para infectar una amplia gama de hospedadores invertebrados y vertebrados. Enterocytozoon bieneusi es el microsporidio más frecuentemente reportado en todo el mundo, principalmente tricrómicaasociado con diarrea crónica y síndrome debilitante en pacientes con sida. Las técnicas dedetección basadas en microscopía y PCR son útiles para el diagnóstico y la identificación deespecies y genotipos, pero estos métodos deben estar estandarizados en cada laboratorio.En este estudio evaluamos técnicas de microscopía y PCR anidada, con secuenciación de losproductos, para detectar E. bieneusi en muestras de heces humanas. Estas técnicas, usadas con-juntamente, podrían ser útiles para su aplicación en el diagnóstico de microsporidiosis intestinaly para realizar estudios epidemiológicos de esta afección en Argentina.

ABSTRACT

Os mecanismos de defesa bioquímica dos anfíbios envolvem secreções cutâneas de moléculas bioativas com atividade antimicrobiana. Este estudo avaliou a atividade in vitro de extratos metanólicos da secreção cutânea de duas espécies de anfíbios da família Bufonidae, Rhaebo guttatus e Rhinella marina, no controle dos patógenos Fusarium udum, Fusarium solani, Colletotrichum truncatum, Aspergillus flavus, Rhizoctonia solani, Macrophomina phaseolina e Calonectria pseudometrosideri. O extrato de R. guttatus inibiu o crescimento micelial de F. udum, F. solani, A. flavus e M. phaseolina em algumas concentrações testadas. O extrato de R. marina inibiu o crescimento micelial de C. truncatum na concentração de 0,5 mg mL- ¹, e inibiu o crescimento micelial de A. flavus nas concentrações de 0,1 e 0,5 mg mL- ¹, que foi semelhante à inibição pelo controle positivo. O extrato de R. marina também diminuiu a produção de microescleródios de R. solani nas concentrações de 0,2 e 0,3 mg mL- ¹. Além disso, os extratos inibiram a esporulação e germinação de conídios em graus variados. A inibição da formação de apressórios em C. truncatum pelos extratos de R. guttatus e R. marina foi de 85%­99% e 63%­100%, respectivamente. Nossos resultados demonstraram que o tratamento com extratos da secreção cutânea de R. guttatus e R. marina apresentou atividade antifúngica sobre os fitopatógenos estudados.(AU)

Resumen: Objetivo: Evaluar el efecto de la combinación de Metarhizium anisopliae y Gliocladium virens, ambos con Aqua Reslin Super, sobre oviposición, eclosión y emergencia de Aedes aegypti. Material y métodos: Se realizaron evaluaciones para determinar el efecto de los tratamientos impregnados en papel filtro y expuestos dentro de recipientes de plástico sobre la oviposición, eclosión y emergencia de Aedes aegypti. Resultados: Los resultados indicaron que las combinaciones hongo e insecticida no afectaron el comportamiento de oviposición, pero sí la eclosión de los huevos y la emergencia del adulto. Conclusión: Con los resultados se puede concluir que la combinación de hongos + insecticida puede ser una buena opción para aplicarse en sitios de oviposición con miras al desarrollo de una ovitrampa letal.


Abstract: Objective: To evaluate the effect of the combination of Metarhizium anisopliae and Gliocladium virens, both with Aqua Reslin Super, on the oviposition, hatching and emergence of Aedes aegypti. Materials and methods: Evaluations were carried out to determine the effect of treatments impregnated on filter paper and exposed within plastic containers on the oviposition, hatching and emergency of Aedes aegypti. Results: The results indicated that the fungus and insecticide combinations did not affect the oviposition behavior, but if the hatching of the eggs and the adult's emergency. Conclusion: With the results it can be concluded that the combination of fungi + insecticide can be a good option to be applied in oviposition sites with a view to the development of a lethal ovitrap.

Abstract Beauveria bassiana is a promising fungus for the biological control of insect pests. The growing costs of conidia production have raised the need to ascertain the efficiency of some low cost substrates. The aim of this study was to analyze the potential use of different raw substrates without nutritional supplement for B. bassiana conidiogenesis. Growth and sporulation were evaluated using 30 g of substrate and 0.3 μL of a conidia suspension (1 x 106 conidia/mL). After 10 days of incubation (70 ± 10% humidity and temperature (T) = 29 ± 1 °C), rice (2.00 x 106 conidia/g substrate), algaroba (2.36 x 106 conidia/g), malt A (1.22 x 106 conidia/g) and malt B (1.75 x 106 conidia/g) showed the highest levels of conidia production. The resulting conidia showed insecticidal activity higher than 80% on coconut termites. These new raw substrates may represent viable alternatives for the production of entomopathogenic fungi for use in the biological control of various insect pests.


Resumo Beauveria bassiana é um fungo promissor no controle biológico de insetos-praga. As crescentes despesas na produção de conídios levantam a necessidade de averiguar a eficiência de alguns substratos de baixo custo. O objetivo deste trabalho foi analisar o potencial de utilização de diferentes substratos brutos para a conidiogênese de B. bassiana. O crescimento e esporulação foram realizados utilizando 30 g do substrato e 0,3 µL da suspensão de conídios (1 x 106 conídios/mL). Após 10 dias de incubação (umidade 70 ± 10% e temperatura T = 29 ± 1° C), o arroz (2,00 x 106 conídios/g de substrato), algaroba (2,36 x 106 conídios/g), malte A (1,22 x 106 conídios/g) e B (1,75 x 106 conídios/g), apresentaram maior produção de conídios. Os conídios produzidos mostraram atividade inseticida sobre o cupim do coqueiro acima de 80% de mortalidade. Estes novos substratos brutos podem representar uma alternativa viável para produção de fungos entomopatogênicos para uso no controle biológico de vários insetos praga.

Abstract To accelerate the recovery of degraded environments, it is necessary to use ecological restoration techniques, which require validation according to the ecosystem conditions where are implemented. This work aimed to evaluate soil microbiological attributes under different ecological restoration technologies in a subtropical forest. The study was conducted at UTFPR-DV, southwest of Paraná, in an ecotone between Mixed Ombrophilous Forest and Semideciduous Seasonal Forest and on an Oxisol. In December 2010, a tillage area of at least 17 years old was isolated and the passive restoration, tree planting and nucleation treatments were installed in 40x54 m plots and four replications. In November 2018 the soil was sampled in these plots and in a native forest area as a reference. There were calculated soil organic carbon content (OCC) and microbiological attributes such as microbial biomass N and C (NMIC and CMIC), basal respiration, fungal spore content and the metabolic (qCO2) and microbial quotient (MICq). It can be concluded that nucleation technology can restore soil microbiological attributes but has not yet reached the conditions of a natural environment. Passive restoration is not a good technology for restoring soil microbiological attributes. The higher contents of CMIC, NMIC, OCC, MICq and fungal spores in the soil under native forest compared to ecological restoration technologies indicate that eight years of adoption of these techniques have not yet been enough to fully recover soil microbiological activity.

In the last decades, the use of entomopathogenic fungi for the control of pest insects has increased globally, however a key step to achieve the successful application of fungal propagules as biocontrol agents depends of various factors, inoculum production being one of the essential stages for these microorganisms to be used in pest management programs. The objective of this study was to evaluate theproduction of conidia by solid fermentation and biphasic culture in different sporulation matrices for four native isolates and a strain of Isaria fumosorosea that have been shown potential for the control of various insects at the laboratory level. The experimental design was completely randomized, two propagation methods were used with six treatments and ten repetitions per treatment; the data were analyzed using an analysis of variance and the means were compared using the Tukey test (p≤ 0.05). In the production by solid fermentation the minimum value of 5.30 × 105 conidia g-1 was registered with the pericarp of peanut and the maximum of 2.35 × 107 conidia g-1 in the corn grain; in the biphasic culture the minimum of 7.60 × 105 conidia g-1 was observed in birdseed and the maximum of 2.07 × 107 conidia g-1 in rice. The differences were significant (p ≤0.05) in the production by method and by substrate, in solid fermentation 6.84 × 106 conidia g-1 and in biphasic culture 8.85 × 106 conidia g-1. In the production by substrate, the rice showed 1.75 × 107 conidia g-1 and the lesser canary seed concentration (7.80 × 105 conidia g-1). The average production per isolate and / or strain was of the order of 106 and significant difference was registered (p ≤ 0.05) among the fungi, the isolate HIB-9 showed concentration in average higher in the production (7.90 × 106 conidia g-1) and the isolated HIB-19 was the least effective, with only 1.08 × 106 conidia g-1. The results obtained show marked differences between the isolates with respect to their capacity to use the different substrates used as propagation media to obtain conidia of I. fumosorosea.


Nas últimas décadas, o uso de fungos entomopatogênicos para o controle de insetos-pragatem aumentado globalmente, porém um passo fundamental para o sucesso da aplicação de propágulos fúngicos como agentes de biocontrole depende de varios fatores, sendo a produção de inóculo um dos estágios essenciais para esses insetos microorganismos a serem usados em programas de controle de pragas. O objetivo deste trabalho foi avaliar a produção de conídios por meio de fermentação sólida e cultura bifásica emdiferentes matrizes de esporulação para quatro isolados nativos e um alinhagem de Isaria fumosorosea que tem demonstrado potencial para o controle de varios insetos em nível laboratorial. O delineamento experimental foi inteiramente casualizado, foram utilizados dois métodos de propagação com seis tratamentos e dezrepetições por tratamento; os dados foraman alisados por meio de análise de variância e as médias comparadas pelo teste de Tukey (p ≤ 0.05). Naprodução por fermentação sólida foi registrado o valor mínimo de 5.30 × 105 conídios g-1 com o pericarpo de amendoim e o máximo de 2.35 × 107 conídios g-1 no grão de milho; na cultura bifásica, o mínimo de 7.60 × 105 conídios g-1 foi observado emsementes de aves e o máximo de 2.07 × 107conídios g-1em arroz. As diferençasforam significativas (p ≤ 0.05) na produção pelo método e pelo substrato, na fermentação sólida 6.84 × 106 conídios g-1 e na cultura bifásica 8.85 × 106 conídios g-1. Naprodução por substrato, o arroz apresentou 1.75 × 107conídios g-1 e a menor concentração de sementes de canário (7.80 × 105conídios g-1). A produção média por isolado e / ou cepa foi da ordem de 106 e registrou-se diferença significativa (p ≤ 0.05) entre os fungos, o isolado HIB-9 apresentou concentração em média maior na produção (7.90 × 106conídios g-1) e o isolado HIB-19 foi o menos efetivo, com apenas 1.08 × 106 conídios g-1. Os resultados obtidos mostram diferenças marcantes entre os isolados quanto à capacidade de utilização dos diferentes substratos utilizados como meio de propagação para obtenção de conídios de I. fumosorosea.

ABSTRACT

Cladophialophora is characterized by simple hyphomycetes with brown hyphae that give rise to branched chains of pale brown conidia and shows affinities with the Herpotrichiellaceae. A fungal strain belonging to the genus Cladophialophora was isolated from soil in Daegu, Korea. This strain produces numerous greenish to dark black lanose aerial mycelia with hair like structures. It is morphological similar to C. chaetospira, C. inabaensis, and C. multiseptata; however, the conidiophores and conidia sizes of the newly isolated strain (KNU16-032) are clearly different from them. The novelty of the strain was also confirmed based on phylogenetic analysis using the data sets of the internal transcribed spacer region of and the partial sequence of 28S ribosomal DNA region along with the cultural characteristics. Because morphological observations and phylogenetic analysis indicated that the strain is distinct from previously known Cladophialophora species, we propose this species as a new species Cladophialophora lanosa sp. nov., and provide the detailed descriptions in this study.

ABSTRACT

A new species of Alternaria causing leaf spots on the rubber tree (Hevea brasiliensis) in Yunnan, China, was isolated, examined, and illustrated. Morphologically, it belongs to the section Porri of Alternaria, which produces relatively large conidia and a simple or branched, filamentous long beak. It is, however, characterized by conidiophores gradually enlarging near the apex into a clavate conidiogenous cell and long ellipsoid to obclavate, smooth-walled conidia with a long filamentous beak. Molecular phylogenetic analyses based on ITS rDNA, GAPDH, and TEF1-alpha sequences demonstrate that the phytopathogen falls in the clade of the section Porri, being most closely related to A. sidae, A. sennae, A. deseriticola, A. cyamopsidis, A. rostellata, A. nitrimali, A. crassa, and A. thunbergiae.

ABSTRACT

Many of the fungicides and antibiotics currently available against plant pathogens are of limited use due to the emergence of resistant strains. In this study, we examined the effects of diphenyleneiodonium chloride (DPIC), an inhibitor of the superoxide producing enzyme NADPH oxidase, against fungal and bacterial plant pathogens. We found that DPIC inhibits fungal spore germination and bacterial cell proliferation. In addition, we demonstrated the potent antibacterial activity of DPIC using rice heads infected with the bacterial pathogen Burkholderia glumae which causes bacterial panicle blight (BPB). We found that treatment with DPIC reduced BPB when applied during the initial flowering stage of the rice heads. These results suggest that DPIC could serve as a new and useful antimicrobial agent in agriculture.

ABSTRACT

Actin filaments play an important role in fungal life processes such as growth, development and cytokinesis. The expression vector pSULPH-Lifeact-mCherry of fluorescent mCherry-labeled actin was transferred into Verticillium dahliae Kleb. wild type V592 by the genetic transformation system mediated by Agrobacterium tumefaciens to obtain the stable fluorescent labeled actin strain V592/Lifeact-mCherry. Then we detected its biological phenotype and the dynamic changes of actin fluorescence during the process of spore germination, mycelial growth and development. There was no significant difference in the colony morphology, colonial growth rate, sporulation and germination rate between the fluorescent labeled actin strain and the wild type. The actin fluorescence signal was observed at the tip of the conidia and hyphae and the septum clearly. Actin participated in the formation of the contractile actomyosin ring (CAR) during cytokinesis by observing the dynamic behavior of the actin in the process of hyphal septum formation. The fluorescent labeled actin strain can be used to study the dynamics of actin in fungal development to provide theoretical and practical support for further study of the mechanism of actin in fungal development and pathogenesis.

ABSTRACT

The aim of this study was to investigate the species of conidial fungi associated with leaf litter of Cedrela odorata (Meliaceae), an endangered red cedar species typical of Amazonian terra-firme forests. Conidial fungi were sampled around C. odorata individuals in three forest areas in the municipality of Belém (Pará State, Brazil). A total of 104 species were identified, with 53 new records for the state of Pará, 46 first records for the Brazilian Amazon, including new records for Brazil (Cordana abramovii), for South America (Acarocybiopsis cubitaensis, Xylocladium claviforme) and for the Americas (Dactylaria biguttulata). A review of species of conidial fungi reported on C. odorata is provided, indicating its distribution in Brazil. For each new record in Brazil, South America and the Americas we present a description, illustrations, geographical distribution and taxonomic comments.


O objetivo deste estudo foi investigar as espécies de fungos conidiais associadas com o folhedo de Cedrela odorata, uma espécie de cedro vermelho ameaçada, típica de florestas de terra firme da Amazônia. Fungos conidiais foram amostrados no folhedo ao redor de indivíduos de C. odorata em três áreas florestais no município de Belém (PA). Um total de 104 espécies foi identificado, com 53 novos registros para o Estado do Pará e 46 que representam o primeiro registro para a Amazônia brasileira. Dentre estas, foram encontrados novos registros para o Brasil (Cordana abramovii), para a América do Sul (Acarocybiopsis cubitaensis, Xylocladium claviforme) e para as Américas (Dactylaria biguttulata). Uma revisão das espécies de fungos conidiais relatadas em C. odorata é fornecida, indicando sua distribuição no Brasil. Para cada novo registro no Brasil, América do Sul e Américas apresentamos uma descrição, ilustrações, distribuição geográfica e comentários taxonômicos.

ABSTRACT

Los hongos micorrícicos arbusculares (HMA) de los ambientes áridos y semiáridos son importantes para el desarrollo de las plantas que crecen bajo estrés biótico y abiótico en áreas naturales o en agroecosistemas. Existe poca información sobre la diversidad temporal de los HMA en plantas perennes de ecosistemas áridos en el norte de México. En este estudiose evaluaron la colonización micorrícica y la diversidad temporal de los HMA en la rizosfera de Larrea tridentata, planta perenne de amplia distribución en el Desierto Chihuahuense. Se obtuvieron muestras de la rizosfera y de raíces de 15 plantas, en 3 fechas de muestreo del año 2015. Se encontró un total de 17 especies de HMA, distribuidas en 12 géneros y 7 familias dentro del phylum Glomeromycota. La especie más abundante fue Funneliformis geosporum. Esta especie pertenece a la familia Glomeraceae, la única que estuvo representada con varios géneros en L. tridentata. El mayor porcentaje de micorrización se presentó en febrero, con un 83,22%, en septiembre fue del 75,27% y en mayo del 65,27%. El muestreo realizado en febrero presentó el mayor número de especies (16), seguido por el de mayo (15) y el de septiembre (12). El análisis estadístico mostró diferencias significativas en el número de esporas entre los muestreos de febrero y los de mayo y septiembre.


Arbuscular mycorrhizal fungi (AMF) of arid and semiarid ecosystems are important for the development of plants that grow under biotic stress in wild or in agro-ecosystems. There is little information on the temporal diversity of these organisms in perennial plants from arid ecosystems in northern Mexico. On this study, the mycorrhizal colonization and the temporal diversity of AMF in the rhizosphere of Larrea tridentata, perennial plant abundant in the Chihuahuan Desert region were explored. Samples of the rhizosphere and roots of fifteen plants in each of the three sampling dates during the 2015 year were obtained. A total of 17 species of HMA belonging to 12 genera and 7 families within the phylum Glomeromycota in all three sampling dates were found. Funneliformis geosporum was the dominant species belonging to the family Glomeraceae which possess the highest genera number on L. tridentata.The highest mycorrhization percentage was in February with 83.22, followed by September and May with 75.27 and 65.27%, respectively. A maximum of 16 AM fungal species were isolated and identified from L. tridentata rhizosphere in February, 15 species in May and 12 species in September. Statistical analysis showed significant differences between sampling dates in the spores number.

Abstract Fungal infections have become a concern for health professionals, and the emergence of resistant strains has been reported for all known classes of antifungal drugs. Among the fungi causing disease, we highlight those that belong to the genus Aspergillus. For these reasons, the search for new antifungals is important. This study examines the effects of a coumarin derivative, 4-acetatecoumarin (Cou-UMB16) both alone and together with antifungal drugs, and its mode of action against Aspergillus spp. Cou-UMB16 was tested to evaluate its effects on mycelia growth, and germination of Aspergillus spp. fungal conidia. We investigated its possible action on cell walls, on the cell membrane, and also the capacity of this coumarin derivative to enhance the activity of antifungal drugs. Our results suggest that Cou-UMB16 inhibits Aspergillus spp. virulence factors (mycelia growth and germination of conidia) and affects the structure of the fungal cell wall. When applying Cou-UMB16 in combination with azoles, both synergistic and additive effects were observed. This study concludes that Cou-UMB16 inhibits mycelial growth and spore germination, and that the activity is due to its action on the fungal cell wall, and that Cou-UMB16 could act as an antifungal modifier.
